Computer-aided design
AutoCAD uses a specific graphic style, developed for its purpose. Unlike other CAD systems, it does not use the Microsoft Windows graphical user interface (GUI) as its default interface and instead uses a non-graphical point and click method. Adriamycin (ADM) is one of the most commonly used chemotherapeutic agents for treating a variety of cancers. Despite the clinical success of ADM, it has a certain degree of toxicity and can result in the emergence of drug-resistant tumor cells. Considering the multiple cellular signaling pathways that are targeted by ADM and the reported synergistic effects of combining ADM with different signaling inhibitors, it is reasonable to hypothesize that combinations of ADM with different inhibitors of specific signaling pathways may be a good strategy to enhance the efficacy of ADM. In this study, we evaluated whether co-targeting EGFR with ADM may enhance the cytotoxic activity of ADM. We found that EGFR expression in human breast cancer cell line MCF-7 significantly enhanced ADM-induced DNA damage, oxidative stress, cell cycle arrest and apoptosis. Our data further showed that while cytotoxic activity of ADM was almost completely abolished by co-incubation with EGFR siRNA, ectopic expression of EGFR significantly suppressed the cytotoxicity of ADM. We further found that combined treatment with EGFR inhibitors gefitinib and cetuximab enhanced ADM-induced apoptosis in MCF-7 cells. The combination also enhanced ADM-induced DNA damage and ROS production.
